hi karin, prob easier to arrange a lift from phuket straight to kanchanaburi(river kwai)easy enough from bangkok if yuo wanna stop off, plenty budget accomadation in kanchanaburi, and best to arrange day trips when you arrive plenty of excursions available from guesthouses etc, pretty inexpensive too, you wont regret going to kanchanaburi with the river kwai, war museum, erawan waterfalls,tiger temple ,loads to see and do have a great time enjoy your hols, and dont forget to pack your chucklemuscles.
